procedure for preparation of microbiological media.

      1.0       Objective
     To lay down a procedure  for preparation of microbiological media.
      2.0        Responsibility
      Microbiologist/QC-Executive.

      4. 0        Procedure
4.1        Take clean glass conical flask.
4.2        Weigh the media as per instructions on the pack as per requirement.
4.3        Add purified water as per requirement in conical flask.
4.4        Check pH at 25°C before of solid and liquid media.
4.5        Adjust the pH value by adding 1N or 0.1N Hydrochloric acid (HCl) or sodium hydroxide (NaOH) solution in order to get the specified pH value.
4.6        Dispense the media as per requirement.
4.7        Close the conical flask with cotton plug.
4.8        Place the flask in the autoclave for media sterilization as directed by the manufacturer and sterilize the media by using autoclave at 15 lbs / inch² for 15 minutes.
4.9        Operate the autoclave as per SOP.

4.10        Enter the details of media consumption in the media preparation record.
